Résumé

Pour contribuer à la lutte contre les mycoses opportunistes à fortes recrudescences suite à l’avènement du VIH/SIDA, notre équipe de recherche a testé l’action de 10 extraits issu des écorces de Terminalia mantaly H.Perrier (TEKAM1) : 2 extraits totaux (aqueux et hydro-alcoolique), six extraits issus de partition liquide/liquide et 2 extraits issus de dégraissage à l’hexane au soxhlet. Ces extraits ont été testés sur la croissance in vitro de Candida albicans, une souche très impliquée dans les mycoses opportunistes. Ils ont été incorporés à la gélose Sabouraud selon la méthode de double dilution en tubes penchés.

Après 48 heures incubation à 30°C, la souche testée est sensible à tous les extraits selon une relation dose-réponse. Le résidu non soluble dans l'hexane T4-2 (CMF = 24,37 μg/ml, CI50 = 15 μg/ml) est l’extrait le plus actif. Par contre, l’extrait dichlorométhanique T3-1 a montré l’activité antifongique la plus faible (CMF=780 μg/ml, CI50=560 μg/ml).

Ainsi l’utilisation du solvant hydro-alcoolique (70% d’éthanol) suivie d’un dégraissage est la meilleure voie qui permet de mieux concentrer le principe actif de TEKAM1.

Abstract

To support the fight against opportunist mycosis which are wildspread because of the advent of HIV, our research team tested the action of 10 Terminalia mantaly (TEKAM1) extracts from stem bark: 2 crude extracts (aqueous and hydroalcoholic), 6 extracts from liquid/liquid partition and 2 extracts obtained by the removal of oil in the soxhlet. These extracts have been tested in vitro against Candida albicans, a strain very often met in opportunist mycosis. These extracts have been incorporated to the Sabouraud gelose according to the method of double dilution in leaned tubs.  

After a 48 hour-incubation at 30°C, the tested strain is sensitive to all extracts according to a measure and effect relationship. The residue of the removal of oil T4-2 (FMC = 24,37 μg/ml, CI50 = 15μg/ml) is the most active extract. On the contrary, dichloromethanic extract T3-1 showed the lowest antifungal activity (FMC=780 μg/ml, CI50 = 560 μg/ml).  

 Therefore, using the hydro-alcoholic solvent (70% ethanol) followed by removal of oil is the best way to obtain an optimally concentrated active ingredient from TEKAM1.
